The Leiden mutation on factor V induces resistance to active Protein C.
This mutation leads to the replacement at position 506 of an arginine by a glutamine, which affects the site of cleavage of factor V by PCa.
Factor V then loses its role as a cofactor of the C protein system, a coagulation inhibitor system.
Factor V retains its procoagulant properties.
